I believe you can fit the lens & cam separate. One of the things I like is the depth and squareness of the case (& weight). You can fit 2cams, lenses with hoods ready to shoot.
The fact that it has a transparent inside pocket helps me in my senior moments when equipping for different shoots. I can see what I have without having to open pockets.
I use the computer pocket for a 1' fold-able bench (from Bed, Bath etc). With it I can shoot over 6' fences at tracks and use it as a seat when shooting polo/surfers (its low enough to raise my knees that support elbows for long lenses
Not exactly sure how long the lens is but I know my 400 2.8 fits great in the Think Tank Glass Taxi (not with a body attached though) and it also fits in my Airport International and it can be attached to a body when in that bag. I'd recommend looking at the offerings from Think Tank as they have awesome stuff and stand behind their stuff and if there is any issue with it they make sure to get it rectified, even if it is not their fault. I absolutely love that company and their products.
