Since you're asking, I'm going to assume you're a little green, is that true? I don't mean that as a negative to you at all, but I think it's important to know before recommending a camera. I'm also going to assume you don't have an investment in lenses.
My opinions:
5D cons - You're going to hate the frequency of sensor cleanings. Once you get over that initial irritant, and become efficient at cleaning, it's no big deal. Still, it's a deal breaker for some. This camera is a bit behind in modern features, some of which the D300 has. The AF may leave you a little frustrated, unless your expectations are realistic, simply consider this a portrait/landscape camera. It will do sports decently with center point AF, and giving the camera a moment to track. The body is not sealed very well, there must be some gaping holes for all that dust to make its way to the sensor!
5D pros - Excellent image quality, and will produce great files with decent glass. Since it's full frame, the 24-105(or 24-70), is likely to be your new best friend. You could loctite the 24-105 to my 5D, and I'd be pretty darn happy with that. Canon also has a much better lens selection in my opinion(with one caveat I'll mention with the D300).
D300 pros - The D300 is a significantly more modern camera, and is just a great all around performer. The AF is noticeably better than the 5D. Live view, some people don't care about it, but I find it pretty handy for macro work. The D300 also has much better weather sealing than the 5D. You can also use a ton of old nikkor gems, they will all be manual focus(all those old Nikon lenses can be used on the 5d too). There's many more modern features, I would recommend you compare the two over at dpreview and see if any strike you as important. The D300 has a way better LCD, though that's something I've never been very concerned about, maybe it's important to you?
D300 cons - Ultimately the image quality will be less than the 5D, though you probably wont notice until you start printing big. I would say the ISO comparisons are close enough to be a non issue, though many don't like the flat smudging that the Nikon does.
As an all around camera, without a consideration for AF lenses, I think the D300 is a better camera hands down. From a strictly IQ perspective, the 5D is the better camera. The selection of lenses alone makes me shoot Canon. I don't like carrying around fat 2.8 lenses, and Canon just kills it in the f/4 category. The 24-105, no Nikon equivalent. The 70-200/4, no Nikon equivalent. The 400/5.6, no Nikon equivalent in AF. Any f/1.2 AF lens? Nope, nada, none. If you only see a kit lens and a 50/1.8 in your future, I would go with the D300. More important than the hardware, is your creativity, have fun with whatever you choose!
