David at Astrojargon has used the Canon SDK to provide information about DIGIC III and DIGIC IV cameras in his program EOSInfo. It is available for Windows only.
Note that the 5D2 must be power cycled (turn it off and then back on) if you were using the camera right before wanting to get the number of actuations. http://astrojargon.net/EOSInfo.aspx
If you run Macintosh then you can use 40D Shutter Count, also by David at Astrojargon. It will work for the 5D2. The difference between EOSInfo and 40D Shutter Count is that 40D Shutter Count only gives the number of actuations and not any other information. If you were using the 5D2 then you still need to power cycle it before taking the reading so that it includes the most recent actuations. http://astrojargon.net/40DShutterCount.aspx
If you run Linux then you need a friend with a different OS. I've had no luck finding EOS utilities for Linux.
