Not an easy question to come up with a simple answer. Every side has its pros and cons I would say. Leica has great reliable things, great lenses, but what to do with a M9 if I need 400 mm focal length? Nikon is currently leading (in my opinion at least) in the full frame field especially with the D800, but I would miss some lenses I really like in Canon land (like the 135/2, some other fast lenses and tilt-shift lenses). Regarding all this, it would be a tough choice right now to vest in a new camera system from the beginning. Since in my experience glass is more important than camera, I would likely stick with Canon and go with a new 5D III (but since I currently already have two 5D II cameras, this is out of question for me to upgrade to the 5D III since I don't see the benefit here for me).
Tough question! It implies we are going to be restricted to one body and lens for all our photo usage. For me, that means a versatile lens, not something super specialized like long telephoto (even a 70-200 or 100 would be too long for most of my shooting), or odd UWA. I need something int he middle range.
The body makes me lean toward the highest IQ and rez in a practical package, not a medium format.
If I stayed with Canon, that would probably mean a 5D3 (although I love 1D bodies! Just wish the 1DX had 24MP or more.) and the new 24-70L II (the 28-300L is amazing, and I'd consider that too -- I have that lens already, but 24mm is very useful).
I don't think anyone can argue that today Nikon has the best imaging with the D800E, so I'd be drawn that way if I really had to start over. The fly in the ointment is the lens selection and features. Offhand, I'd say coupled with a Zeiss lens, but I can't think of a single prime that would be versatile enough for me.
Since I'm most familiar with Canon operation, I'd go 5D3 with grip, 24-70L II.
Now, if we could just talk Canon into a 30 MP 1DX Mark II with 20-80/3.5 L IS, I'd be set!
I am really happy with my 200 1.8L and 5DII, but if pressed I suppose it would be 1DX and 50L. I wouldn't buy a 1DX however since it is 7K and I am not getting rentals on it like I do my lenses.
