If I'm on vacation, pictures are secondary to the experience. Nobody will wait for me to change lenses. I would happily bring one camera and one *small* lens, or an iPhone.
But yes, if you're willing to carry the bulk and weight, the 28-70/2 GM is a dream lens.

I sometimes berate myself for holding on to too many lenses that I don't often use. But, as you point out, well-chosen lenses all record images with a different look, and those differences are very valuable. I do like the rendering of my Sony 24-70 f2.8 II GM zoom (better, in fact, than what I have seen of the Sony 28-70 f2.0 GM zoom), but the character and rendering is quiite different fron a Loxia 21mm or 50mm, or a Batis 40 or 85, or a Sigma 45/2.8.

I like the minutae of different lenses, even if only I can see the difference, as that's the point. It's for me. I'm not showing in a gallery.

I still think the Pentax 6x7 body was at the top of cool/beautiful cameras. You could also use the shutter ‘clunk’ as a way for people miles away to find you to rescue you in the dark.
Loved both the 105/2.4 and the 135/4 as handheld portrait lenses; though 55 years ago, I was younger, taller, stronger, more energetic; had non-arthritic hands; was more experimental, and thought I was more hip. Pretty sure it’s size and appearance were 95% of reason i got a couple of jobs at Neiman-Marcus, taking bts at fashion shows. 3% as cool as David Hemmings in ‘Blow-Up’ but 110% of the endorphins. 🙈🙈
I way enjoyed it, but deserve zippo-credit for handling it!

The 28-70 f2 definitely interests me as a way streamline how many lenses I have in my bag, make less decesions abut changing lenses, allow me to get more photos faster, and experiment with some fast focal lengths I have never had primes for. Within reason, I am more concerned about total gear weight than the weight of a single lens.
While this would probably mean my CV lenses will end up in my bag less often, I don't see myself selling them, as I feel like they will still have a place for times when I want smaller lenses, MF, and the CV look.
One thing to keep in mind, though, is 110mm is a lot more reach than 70mm, so you may find yourself lacking reach on the long end.

Shooting film and the quality of medium format was very noticeable compared to 35mm film.

But let me try to comment more or less on everything.
I'm 37 right now and while my back is a bitch and has been for years, as an amateuer powerlifter I'm quite strong. A 1 kg isn't that much of an issue to me - as I've wrtitten in an earlier comment, I also have the Sigma 35/1.2 (1st gen).
Concerning differnt rendering charracteristics of different primes vs one zoom I have to say that I think Loxia 25/2.5, CV 50/2 and CV 110 2.5 are very similar, so this is nothing I worry about.
Besides, I also always bring a fast prime. On my last 3 or 4 trips, this has been the Syoptic 50/1.1. This lens has plenty of character!
It was mentioned that I should work with the lens I have mounted. From a creative standpoint I agree but when you've stopped somewhere on a mountain road and want to capture the layered relief of the mountains, 50mm won't get you far. Conversely, having a 300mm lens mounted won't do well in a dark cave.
I am totally aware 70mm is noticeably shorter than 110mm. I have been thinking about this plan for weeks and have gone through all the options. Part 2 of my "plan" is to replace the Nikon 300/4 PF ED with the 100-400/4.5-5.6, so that I have an replacement for the magnification I lose compared to the CV 110/2.5.
As for the enjoyment this hobby should bring: this is a two-part thing. I very much enjoy using MF primes but having to stop every few minutes (worst case) to change lenses takes away from that joy. Sometimes having AF would make things easier but I know the Sony 28-70 won't give me the same tactile joy. However, it would allow me to concentrate more on the vacation part. I like to mix it up on vacations. For instance, we went to Paris last May and I used a 28/1.4 manual focus prime 75% of the time, swapping it for a 50/1.4 or 10/5.6 the rest of the time. A few weeks ago we went to the Big Island of Hawaii and there I used a 35/1.4 manual focus prime 75% of the time, swapping it for a 75/2 or 21/1.4 the rest of the time. Our next trip will be to France in a few weeks and this time I’ll take the 24-70GMII along with a 35/1.4 manual focus prime, a 14/4, and possibly a 21/1.4. It’s fun to mix it up a bit.

I’m still a prime shooter, but I’m shooting zooms as well, much more than I used to. Before it was pretty limited to tele zooms.

Depends on how many primes you bring with you to cover that range. That might also require a large bag and also be heavy. (and possibly involve even more lens changes.)
To some extent this is a futile exercise in applying one’s own value system when commenting, which works perfectly when having a conversation with yourself.
In reality, no one strategy is wrong, but no one strategy is right for everyone either. It’s personal preference.
In the end do what makes you happy.

I used the 86mm to 77mm step down with achromat and there was some fall off but definitely usable. 86mm to 82mm step down didn’t seem really affect much so I just left that on (stacked on top of 86mm filter).
IQ wise it does replace all those lens and even matches the Otus 50mm 1.4 ML. The 28-70 GM is the last AF lens I sold before deciding to leave Sony for personal use.
All day handling is easy and well balanced about right at the limit. It also fits in most normal size bags easily.
If given the chance I would test it out at the store with the same body youre using just for a ergos check but I think you'll be happy.

Perhaps. But now the OP is talking about replacing three primes, two of which are quite small and light, with two large heavy zooms. Further, swapping out two large heavy zooms while sightseeing with others is a different task than swapping out smaller and much lighter lenses. And then also, the weight in hand while taking pictures all day is much greater with the two zooms.

Let me clear that up: my initial question was the one from the title but I only didn't mention the 100-400 to not further complicate things and prevent more alternative lens suggestions I didn't ask for.
So what I'm cotemplating is whether or not I should replace the Loxia 25/2.4, CV 50/2, CV 110/2.5 and Nikon 300/4 PF ED with the Sony 28-70/2 and 100-400/4.5-5.6.
That's 2415 g vs 2353 g.
No, I wouldn't need a bigger backpack, but I'd need to reconfigure my dividers.
Lens changes with a lens like the 100-400 are more difficult but it's not like I could pocket the 300/4, so no practical difference I think.

Thanks a lot for answering one of my questions! I think this settles it for me.
Maximum magnification with the Canon 500D would be 1:2.7 at 70mm (which is actually a mere 58mm at MFD) which is very useable and should cover most appllications.
For the 100-400 values are obviously even higher.
