How is the difference in heft feel compared to the 100-400. Feel like you can hand hold for a bit of time without too much strain?

It's definitely heavier than the 100-400 but hand holdable. Better than the Nikon 500 F4 and the Sigma 150-600 Sports and the 60-600 Sports I had.
