Sure you can.
Its just simpler to see if your Intel iGPU is functioning correctly in CS6 without the need of removing your Radeon card, as long as you set your hardware acceleration in CS6 to Intel HD 4000 instead of Radeon or even disabling that option in CS6 for testing purposes.
