The 400 DO and DO II indeed use diffractive optics as do the Nikon PF lenses.
The Canon 800/5.6 are, however, conventional refractive optics, not DO. The reason the RF 800/5.6 is so short compared to its predecessor is that they went with a design which uses a teleconverter-like rear group on a 400/2.8. It allows close focus and a more compact overall design but does not use DO.
