Something like the Canon EF 70-200 L (f/2.8, /4) or EF 80-200/2.8 L would fit the bill (internal focusing and AF). So would a Sigma 120-300/2.8 EX, but that might be a bit too large, for both size and price.
I can't think of any 'alternative' manual focus zooms in this range that have internal focus, but you could buy two or three really nice alt primes, and still save money compared to buying a used L-series lens.
As far as zoom lock goes, I find a piece of gaffers tape will lock an extending zoom at any position you like.
