The way I understand it, exposure compensation is a bias for the camera's automatic exposure measurement. If you set all the exposure variables manually (ISO, aperture, shutter speed), this has no meaning.
ISO is the sensor light sensitivity setting, which is one of the exposure variables, next to aperture and shutter speed. If you are in one of the automatic modes, changing ISO will not affect exposure. Just like changing aperture in A mode, or changing shutter speed in S mode, doesn't affect exposure. Here you need the exposure compensation setting to affect exposure in those modes.
Bang on. EC has no effect when in full manual mode with a hard set ISO. It's only to push/pull exposure when in a mode like Av or Tv or when shooting M with auto ISO.
