Hey Bob,
When shooting the Moonbow of Yosemite Falls your settings will end up being similar to if you were shooting stars, but in the end, just watch your histogram. Usually for night shots to still feel realistic, you want them to be one to two stops underexposed on your histogram, so just adjust your exposure to get that.
The shot here is a pano I shot of the Moonbow a few years ago. It's at f5.6, 25 sec shutter speed and at ISO 2500. Some generic star shot settings. You can start there for an idea, but again, look at your histogram to adjust the exposure.
As for focusing, just use your autofocus on the D800E to gain focus, then turn it off. The D800E can easily focus at night, just focus on a contrast point like the edge of the waterfall against the granite wall, or the granite wall against the sky.
One more note, a polarizer can help to pull more detail out more detail of the Moonbow so try that too on a few shots and see how it looks.
