You'll probably find tons of content in a web search about the uses of rear-button focus.
But to summarize, the primary benefit to decoupling autofocus from the shutter release is that it puts you in charge. You can focus, recompose, then fire off several frames without worrying about the camera refocusing where you don't want it to. This is particularly useful on cameras like the 5D Mark II, where you have solid center point AF, but outer focusing points that cover a limited portion of the frame or can be unreliable in difficult lighting.
Pressing the shutter button always activates the meter. The two options shown let you choose between metering with or without AE lock. If you are familiar with AE lock already, then deciding should be straightforward. (Some photographers love it, but I never use it, preferring to use exposure compensation or manual mode instead.) Experiment to see what you prefer.
