I will be doing this adjustment with my lens after Christmas
I have never done it before
I do know:
Tripod
Av mode at wide open aperture
What I don't know
What ISO?
And do I take 2 or 3 photos and compare before making + - adjustments or just 1prior and after adjustments?
Single point or Spot single point?

I will be using a Datacolor SpyderLensCal SLC100

This is similar to LensAlign. You want to use the following:

a tripod
shutter release or use timer (not the 2 second one either)
Distance = 25-50 x the lens focal length
Use a low ISO (400-800)(don't know which camera your doing)
Defocus and allow camera to acquire focus
Take one shot at "0" and look to see if it's back or front focusing (personally, I do this on the computer and not the camera screen), adjust accordingly, take another shot and repeat.

Canon 7D MkII and several lens
70-200 2.8 IS Mk1 and then also with Tc1.4
85 1.8
50 1.4
135 F2
17-55 2.8 IS

I may be completely wrong but in my experience, the focus varies quite a bit with different light levels (for example bright outdoors versus at night indoors). My Canon 5D3 manual alludes to this also. So I make a point to calibrate using the light conditions, camera settings, etc. that I will most likely use the combo in. So far I've been fairly successful (or lucky or both).

Yes, you are correct^^^. I forgot to mention lots of light. I try and perform this outdoors when possible, but when not possible, I use some 1000w or higher bulbs on the target or flash.

You should have no trouble calibrating those lenses outdoors since they are short focal lengths.

Methodical and I use the same system...quick and accurate after you've done it a few times.

Don't use spot. Use single point.

Rather than just diving in and AFMA each lens why not use the equipment for a while and see from your images whether AFMA is required. My most recent camera, 6D, has required no adjustments with my tele-lenses. My 7D did, but once again I did this with a bit of experience behind the adjustment. I did not needed to AFMA any of my wider lenses.
