A question, once an adjustment has been made and recorded in-camera, does the adjustment automatically go into effect when the lens is installed or is it necessary to turn on auto fine tune?

AF Fine stays on until you turn it off through the menu system. If on, it automatically adjust AF per saved adjustment. Where the default or specific by lens.

So, if I understand this... as long as the af fine tune is on via the menu, the camera will make appropriate adjustments for each calibrated lens/lens combination that is placed on the camera.

There are exceptions to this. In the case of Zeiss lenses for example only ONE lens is recognized.

I don't understand using AF Fine tune for a manual focus lens?

Nikon cameras will only recognize one Tokina lens, so if you have more than one you have to choose which needs AF Fine Tune more.
